Anti-inflammatory and Antinociceptive Effect of the Water Soluble Fractions of Bee Venom on Freund¡¯s Adjuvant Produced Arthritis in Rats

Abstract
We previously demonstrated that the acupoint injection (Zusanli, ST36) of whole bee venom (BV) and its water-soluble extract (BVA) had more potent antinociceptive and anti-inflammatory effect on Freund¡¯s complete adjuvant (FCA)-induced arthritis as compared with that of non-acupoint injected animals. In present study, we further separated BVA to three fractions (i.e. BVAF1;>20kDa, F2;10-20kDa, and F3;<10kDa) according to the molecular weight to elucidate the most effective component in BVA. Rheumatoid arthritis was induced by FCA injection into the plantar surface of the right hind paw. The arthritis induced inflammation and nociception were observed in the contralateral left hind paw from 12 days after the FCA injection. Subcutaneous injection of BVAF1(0.4 mg/kg) and BVAF3(1 mg/kg) into the Zusanli acupoint dramatically suppressed both paw edema and radiological change (i.e. new bone proliferation) caused by FCA injection. In addition, both BVAF1 and BVAF3 injection significantly reduced arthritis-induced nociceptive behaviors (i.e., nociceptive scores for mechanical hyperalgesia and thermal hyperalgesia). Finally, BVAF1 and BVAF3 treatment significantly suppressed FCA induced Fos expression in the lumbar spinal cord at 3 weeks post-FCA injection. In contrast, BVF2 treatment did not show any anti-inflammatory or antinociceptive effects on FCA induced arthritis. In conclusion, the results of the present study demonstrate that BVAF1 and BVAF3 fractions may be the effective fractions of water soluble BV component that are responsible for the BV induced antinociception and anti-inflammatory effects.
